FORMULATION OF DILTIAZEM PELLET BY LAYERING METHOD IN FLUID-BED MACHINE

Abstract:
In this research, the pellet containing diltiazem HCl has been initially
studied by layering method in fluid-bed machine. The formulation of
the pellets included avicel, lactose, binders and diltiazem. Pellets
containing ditiazem was prepared by layering drugs and excipients on
the inert core which was prepared in rotary processor. The diltiazem
pellets were coated with the layering drugs containing ditiazem in
fluid-bed machine to obtain pellets. The prepared pellets were
evaluated for drug content, particle size, subjected to Scanning
Electron Microscopy (SEM) and Differential Scanning Calori metry
(DSC), and evaluated for in vitro release. Diltiazem pellets achieved
good sphericity, low friability, narrow particle size distribution,
smooth surface, high efficiency and be able to control release the drug
well. From the research results, we have developed a formula for
preparing diltiazem pellets by the fluidized bed coating method from
inert cores and standards for pellets as a semi-finished product for
capsule or tablet filling or extended-release formulation
